I think certain characteristics can be stereotyped into the gay ballpark, but in essence, anyone can be anyone, so there is just no telling how to be 100% sure. I'm obviously very camp, which stereotypes me into the gay catagory, but I could be just camp.
To be honest, I don't give a shit. It's none of my business who sleeps with who. So I have no need to guess. If someone tells me, that's fine, but in my experience, that is the only way to truly prove someones sexuality as gay or straight.

As Seth said, certain characteristics can depict people to be a certain orientation. Being camp (or feminine for people who aren't British) is a very obvious one, but doesn't always prove to be correct.
A more technical way of viewing it , would be through sex pheromones. These pheromones act as chemicals that only affect things of the same species. In this case pheromones are secreted by humans (Males: underarm - Females:Head) and it triggers a receptor found on another human (nose).. pheromones are used to attract one another. Sometimes you'll find a greater initial attraction to someone if you both bear the right pheromones and receptors. Think of it like a key and a lock. It is proposed that gay men secrete different pheromones and have different receptors than straight men/women. Its just a theory I think, but i could be wrong. Im not sure how much research has been done on the topic, but it seems very viable. I suppose that could be a type of gaydar

gaydar is nothing but paying close attention. the reason some know when others are gay and say its their gaydar it is just them paying attention, i for one with my ADD pay attention to EVERYTHING so i have a hightend gaydar but a really close friend can tell if someone is gay even before the person has come out to his or herself... scary

I believe that gay people can be mistaken for straight people with out a thought going through a gay person head. I believe that gaydar is based on general gay stereotypes that have been passed down from generation to generation from society. I wish I was able to tell who's gay and who's not. It would help me find relationships.
